摘  要:选取3个骨干玉米自交系丹988、齐319、郑58幼胚为材料,利用正交试验设计L9(34),研究了基因型、幼胚大小、2,4-D浓度和AgNO_3浓度4个因素对胚性愈伤诱导率的影响。结果表明:各因素对愈伤诱导结果的影响大小依次为基因型>幼胚大小>2,4-D浓度>AgNO_3浓度,最优组合为"齐319"、幼胚为1.5~2.0 mm、2,4-D浓度为2 mg/L和AgNO_3浓度为8 mg/L,愈伤组织诱导率为60.58%。用农杆菌介导法将植物超表达载体p CAMBIA3301-ICE1-Bar转入玉米自交系"齐319"中,得到的转化植株经除草剂basta初步筛选后,通过PCR检测,获得了5棵T_0代阳性植株。

The embryos of three major maize inbred lines Dan988, Qi319 and Zheng58 were select- ed as gene receptors to explore the effects of genotype, size of embryo, concentration of 2,4-D and concentration of AgNO3 on the induction rate of embryonic callus by L9(34) designed orthogonal ex- periment. The results showed that the effects of each factor on the embryonic callus induction rate was genotype〉 embryo size 〉 2,4-D concentration 〉 AgNO3 concentration and the optimum condi- tions were maize inbred line Qi319, embryo size 1.5-2.0 mm, 2,4-D concentration 2 mg/L and AgNO3 concentration 8 rag/L, under which the embryo callus induction rate could be as high as 60. 58%. The plant overexpression vector PCAMBIA3301-ICE-Bar was transferred into maize inbred line Qi319 by agrobacterium and 5 TO transgenic positive plants were obtained through the prelimina- ry screening by herbicide basta and the followed detection by PCR.
